PEGylated sequence-controlled macromolecules using supramolecular binding to target the Taspase1/Importin α interaction

چکیده

PEGylated sequence-controlled macromolecules using supramolecular binding motifs effectively disrupt Taspase1 interaction with Importin α in a concentration-dependent manner, thereby exploiting novel inhibition mechanism for this protease.

Predicting an Aptamer’s Target Binding Affinity Using its Nucleotide Sequence

Modern molecular and cellular biology depends on markers that can very selectively bind to a target molecule. Unfortunately, the process of finding such a marker is generally slow, through trial and error, and often in vivo. Aptamers (short nucleotide sequences) are a promising type of marker that can be generated and evaluated synthetically. Currently, the process of aptamer selection involves...

Controlling monomer-sequence using supramolecular templates

The monomer sequences in biopolymers such as nucleic acids and proteins are strongly linked to their function, and even small errors can yield non-functional macromolecules, or worse. Nature therefore expends significant resources on the (nearly) flawless synthesis of these polymers. For example, DNA is replicated using DNA polymerases, which achieve error rates as low as 1 in 10 nucleotide add...

The PUMILIO-RNA interaction: a single RNA-binding domain monomer recognizes a bipartite target sequence.

Translational repression of hunchback (hb) mRNA in the posterior of the Drosophila embryo requires two copies of a bipartite sequence, the Nanos Response Element (NRE), located in the 3' untranslated region of the mRNA. The PUMILIO (PUM) protein is thought to bind the NREs and thereby repress hb translation. The RNA-binding domain of PUM defines an evolutionarily conserved family of RNA-binding...

Irreversible binding of chlorinated ethylenes to macromolecules

Rats have been exposed in a closed system to the chlorinated ethylenes vinyl chloride and trichloroethylene and to carbon tetrachloride as a reference compound. Data of uptake of the compounds, of urinary excretion of metabolites, and of exhalation after exposure show that the chlorinated ethylenes are metabolized much faster than carbon tetrachloride, probably due to their common ethylene stru...

Small and stable peptidic PEGylated quantum dots to target polyhistidine-tagged proteins with controlled stoichiometry.

The use of the semiconductor quantum dots (QD) as biolabels for both ensemble and single-molecule tracking requires the development of simple and versatile methods to target individual proteins in a controlled manner, ideally in living cells. To address this challenge, we have prepared small and stable QDs (QD-ND) using a surface coating based on a peptide sequence containing a tricysteine, pol...
